CCP-EM Leeds RELION Workshop

In Leeds
Hosted by CCP-EM and the Astbury Biostructure Laboratory (University of Leeds, UK) this one day workshop will focus on the RELION software package and is led by its developer Sjors Scheres (MRC-LMB).
The training will involve a hands-on tutorial of RELION 2.0 for cryo-EM single-particle analysis (http://www2.mrc-lmb.cam.ac.uk/relion). It will cover the entire process from initial micrograph inspection, CTF estimation, (automated) particle picking, 2D and 3D classification, 3D high-resolution refinement, movie-processing, and map sharpening.
Computational facilities will be provided along with refreshments.
Registration is free but not a guarantee of acceptance. Application for registration will close on Monday 15th August 2016, participants will be selected and we will confirm places no later than Friday 19th August 2016. There are 30 places at the workshop.
61st Annual Meeting of the Biophysical Society (with new Cryo-EM Subgroup meeting)

To address the recent surge of interest in the general biophysical community, the Cryo-EM Subgroup was initiated at the 2016 meeting with a program of speakers that were
selected from submitted abstracts. For the 2017 meeting, we have invited

subgroups earlier in the day. Other Cryo-EM sessions at the meeting
include a Workshop as well as a Platform Session with speakers selected

Joint International Conference of the Hellenic Crystallographic Association and the Hellenic Society for Computational Biology and Bioinformatics - HeCrA-HSCBB16

The 8th International Conference of the Hellenic Crystallographic Association (HeCrA) and the 11th Conference of the Hellenic Society for Computational Biology and Bioinformatics (HSCBB) are jointly organized as HeCrA-HSCBB16.Â
HeCrA-HSCBB16 will take place at the Agricultural University of Athens in October 7-9, 2016.Â
The conference will address key questions and issues in molecular structure and interactions. The Conference is the major event both of HeCrA & HSCBB. It is a forum to communicate state-of-the-art research, interact and establish scientific networks and collaborations.
In particular, it will bring together crystallographers with Bioinformaticians and Computational Biologists for the identification of macromolecular targets and small molecules of pharmaceutical and biotechnological interest and study their interactions.
There will be oral and poster presentations in addition to talks from distinguished invited speakers from Greece and the European Union.Â

MX beamline training session

Organized by the Diamond Light Source Macromolecular Crystallography group.
The aim is to provide MX users with sufficient training to be able to operate any of the Diamond MX beamlines efficiently and to get the most benefit from their beam-time. The training will involve hands-on sessions on the suite of five operational MX beamlines (http://www.diamond.ac.uk/mx-home/) as well as optional offline software sessions.

MicroED workshop (second installment of the)

MicroED is a cryo-EM method that uses electron diffraction to facilitate structure determination from sub-micron sized 3D crystals of biological material. MicroED uses equipment and procedures that are standard on many cryo-EM:s, and because of this the technique can be easily adopted in labs and facilities around the world. To encourage the widespread use of MicroED we are offering this workshop to teach microscopists how to perform the technique and adopt it at their own institutions. The first MicroED workshop was held in February of 2016 and was heavily oversubscribed. For this reason we are offering a second workshop.
There is no registration fee for participation, and Janelia will provide on-site housing and meals. Attendees are expected to cover their own travel expenses and must be able to stay for the duration.
Space in the course is limited. The application deadline is August 1, 2016 (11:59 PM EDT)

The workshop will provide hands-on training on practical aspects of 2D crystal data collection on a Titan Krios, and cover in depth the theoretical foundations and the practical image processing of 2D crystal cryo-EM images with the 2DX software package. No previous experience is required.
Topics include:
Basics of 2D electron crystallography
- Introduction to 2D electron crystallography
- Cryo-electron microscopy sample preparation for 2D crystals of membrane proteins
- Cryo-EM data collection of 2D crystals
- Introduction to 2DX
- Drift correction using ZORRO Â
Basic image processing with 2DX
- Determination of the defocus, crystal tilt geometry, crystal lattice
- Crystal Unbending - including movie-mode drift correction of individual unit cells in dose-fractionated image series
- 2D merging and 3D merging with reconstruction via weighted back projection
- Automation of the processing pipeline
Advanced topics in image processing with 2DX
- Approaches for a partial reconstruction of amplitudes and phases inside of the missing cone
- Processing of 2D crystal unit cells with RELION and FREALIGN, under exploitation of neighborhood correlation of certain image parameters
- Advanced usage of the 2DX backend library
- Preparation of final results with quantitative description of statistical values (e.g., resolution) for export to databases (EMDB).
Computing infrastructure will be provided in place and participants are welcome to bring their own data sets and/or computer (OSX or Linux).

The workshop will provide hands-on training on all practical aspects of sub-tomogram averaging, encompassing all stages of the pipeline: tomogram visualization and archiving, particle picking in different geometries, averaging, refinement and classification. The course is intended primarily for post-doctoral researchers and Ph.D. students in structural biology with at least one year’s relevant experience in 3D-structure determination of macromolecules (X-ray diffraction, SAXS, cryo-electron microscopy). The workshop will cover state-of-the-art methodology for data processing with emphasis on integration of different methods to solve structures of macromolecular complexes and ‘difficult’ targets. The courses will cover theoretical and practical aspect of:
- X-ray data processing (diffraction and SAXS)
- Electron microscopy data processing
- Phasing, refinement and validation (structure and data)
- Integration of data from different experimental sources (X-ray diffraction and diffusion, cryo-electron microscopy), structure prediction to solve difficult problem (macromolecule reticent to crystallization, large complexes)
- Ongoing and future developments (methods, techniques and instruments) for integrated structural biology.
The number of participants is limited. 25 candidates will be selected by a committee (see Website for registration details and application). No registration fees for the workshop.
The workshop will be couple to the meeting "Current Challenges in Integrated Structural Biology" (October 3 - 4, 2016, IGBMC). A web site will be open shortly for the meeting. We strongly encourage participants of the workshop to attend the meeting.
This workshop forms part of the Instruct and of the FRISBI Training Course Programme 2016. It also part of the French national PhD program in Integrated Structural Biology (ReNaFoBiS).

While the primary purpose of the course is educational, students may bring a limited number of their own research samples. Prepared standard protein samples will be available to all students for practice.
This course now accepts remote students. These students will receive digital copies of all lectures, tutorials, and sample data, and will have access to a live stream of the course lectures. Remote students will not have data collection time during the course. This option is intended for current MacCHESS BioSAXS users who want a refresher, or for new users who have scheduled beamtime this May but cannot come to CHESS twice.
